Ewing's Sarcoma Treatments: Platinum Compounds
There are a number of chemotherapy agents which are considered platinum compounds: carboplatin, cisplatin, and oxaliplatin. They contain platinum complexes which are believed to inhibit DNA synthesis by forming interstrand and intrastrand cross-linking of DNA molecules. They are cell-cycle-phase nonspecific and are considered to be radiation-sensitizing agents.
Carboplatin
- Carboplatin is frequently used as one of the chemotherapy agents for stem cell rescue; see the Stem Cell Rescue page for the research results for stem cell rescue with carboplatin.
- 2000 abstract indicating that carboplatin with ifosfamide might cause renal tubular toxicity.
- 1994 abstract detailing results of a Phase 2 trial of carboplatin; 1/15 CR for Ewing's. Plus 4/7 of the patients who responded to carboplatin had had cisplatin previously
Cisplatin
- 2001 abstract discussing the overexpression of the HER2/neu oncogene which is associated with tumorigenicity and drug resistance.
- 2000 abstract discussing the SSG IV and IX protocols. The SSG IX protocol employed cisplatin and other chemotherapy agents.
- 2000 abstract discussing Etoposide/Ifosfamide/Cisplatin (VIP) in the treatment of Ewing's Sarcoma unresponsive to standard therapy.
Oxaliplatin
-
Current clinical trial using oxaliplatin in treating young patients with recurrent solid tumors that have not responded to previous treatment. Ewing's sarcoma (as well as other types of sarcoma) patients are eligible.
